Simply clean your plot once, wait 2 to 3 weeks to give the weed seeds time to germinate. Then do a second cleaning to have perfectly clean soil before sowing the flower meadow. After removing the last traces of weeds, give it a good dig to loosen the surface of the soil to create a well-refined seedbed.
